چکیده

Summary Determination of what is the specificity subunits composing a protein complex essential when studying gene variants on human pathophysiology. The pore-forming ?-subunit KCNQ1, which belongs to voltage-gated ion channel superfamily, associates its ?-auxiliary subunit KCNE1 generate slow cardiac potassium IKs current, whose dysfunction leads arrhythmia. Using pharmacology, invalidation, and single-molecule fluorescence assays, we found that fulfils all criteria bona fide auxiliary TMEM16A chloride channel, anoctamin superfamily. Strikingly, assembly with switches from calcium-dependent voltage-dependent channel. Importantly, clinically relevant inherited mutations within TMEM16A-regulating domain abolish modulation, suggesting TMEM16A-KCNE1 current may contribute pathologies. Altogether, these findings challenge dogma regarding complexes questions classification.

AMIGO is an auxiliary subunit of the Kv2.1 potassium channel.

Kv2.1 is a potassium channel α-subunit abundantly expressed throughout the brain. It is a main component of delayed rectifier current (I(K)) in several neuronal types and a regulator of excitability during high-frequency firing. Here we identify AMIGO (amphoterin-induced gene and ORF), a neuronal adhesion protein with leucine-rich repeat and immunoglobin domains, as an integral part of the Kv2....

beta 3: an additional auxiliary subunit of the voltage-sensitive sodium channel that modulates channel gating with distinct kinetics.

The voltage-sensitive sodium channel confers electrical excitability on neurons, a fundamental property required for higher processes including cognition. The ion-conducting alpha-subunit of the channel is regulated by two known auxiliary subunits, beta1 and beta2. We have identified rat and human forms of an additional subunit, beta3. It is most closely related to beta1 and is the product of a...

A distinct three-helix centipede toxin SSD609 inhibits Iks channels by interacting with the KCNE1 auxiliary subunit

KCNE1 is a single-span transmembrane auxiliary protein that modulates the voltage-gated potassium channel KCNQ1. The KCNQ1/KCNE1 complex in cardiomyocytes exhibited slow activated potassium (I(ks)) currents. Recently, a novel 47-residue polypeptide toxin SSD609 was purified from Scolopendra subspinipes dehaani venom and showed I(ks) current inhibition. Here, chemically synthesized SSD609 was sh...

Stoichiometry of the KCNQ1 - KCNE1 ion channel complex.

The KCNQ1 voltage-gated potassium channel and its auxiliary subunit KCNE1 play a crucial role in the regulation of the heartbeat. The stoichiometry of KCNQ1 and KCNE1 complex has been debated, with some results suggesting that the four KCNQ1 subunits that form the channel associate with two KCNE1 subunits (a 42 stoichiometry), while others have suggested that the stoichiometry may not be fixed....

Neto1 is an auxiliary subunit of native synaptic kainate receptors.

Ionotropic glutamate receptors of AMPA, NMDA, and kainate receptor (KAR) subtypes mediate fast excitatory synaptic transmission in the vertebrate CNS. Auxiliary proteins have been identified for AMPA and NMDA receptor complexes, but little is known about KAR complex proteins. We previously identified the CUB (complement C1r/C1s, Uegf, Bmpl) domain protein, Neto1, as an NMDA receptor-associated ...
